Neil had a normal childhood like most children and completed his education at HR College, where he earned a Bachelor of Commerce degree.
However, he was always focused on pursuing a career in the film industry, which led him to participate in acting workshops and training programs. He underwent a four-month workshop with Kishore Namit Kapoor and also trained at Anupam Kher's institute during his vacations.

Neil made his acting debut as a child in Vijay (1988) and Jaisi Karni Waisi Bharnii (1989). He portrayed a younger Rishi Kapoor in Vijay and a younger version of Govinda's character in Jaisi Karni Waisi Bharnii.
Johnny Gaddaar (2007) was his first leading role as an adult. He appeared in the 2009 science fiction thriller Aa Dekhen Zara. He co-starred alongside Bipasha Basu as a struggling photographer who receives a camera from his scientist grandfather, the photographs of which predict the future. Kabir Khan's drama thriller New York, starring John Abraham, Katrina Kaif, and Irrfan Khan, was his big break. His performance received high praise from critics and the audience.
In 2010, Mukesh starred alongside Deepika Padukone in the action drama Lafangey Parindey. His performance as a boxer was praised by reviewers.
In 2011, Neil participated in Vishal Bhardwaj's film 7 Khoon Maaf, a dark comedy-drama that was inspired by Ruskin Bond's short story Susanna's Seven Husbands. The film stars Priyanka Chopra as a woman who kills her seven husbands in an endless search for love, while Neil plays a supporting role in the film.
Neil Nitin Muskes appeared in Abbas-Mustan's Players in 2012 alongside Abhishek Bachchan, Sonam Kapoor, Bipasha Basu, and Bobby Deol.
