The story of Humshakals centres on the lives of three guys with the same name but different personalities. When the plot brings Ashok, Kumar, and Mamaji together for the first time, they are all strangers to one another. The three main characters get into many amusing misunderstandings and comical predicaments.
As the story progresses, it becomes clear that Mamaji is mentally unstable, Kumar is a small-time con artist, and Ashok is a wealthy businessman. The three men wind up at a mental hospital run by the villainous Dr. Shivani due to a twist of fate and a misidentification. Unbeknownst to them, Dr. Shivani has her own hidden agenda and aims to profit off their identities.
Ashok, Kumar, and Mamaji discover their female lookalikes in the psychiatric hospital, who also happen to be female versions of themselves. Due to the fact that the female characters also have the same names as the male heroes, confusion and disorder are exacerbated. Together, they come up with a strategy to leave the facility and reveal Dr. Shivani's nefarious motives.
